In the early Sengoku period, Izumi was contested under Hosokawa rule among various Hatakeyama factions and local samurai, with Miyoshi Nagayoshi eventually gaining ascendancy. The province became a domain of Oda Nobunaga in 1573, and after the Honnō-ji Incident passed to Hashiba Hidenaga's rule under the Toyotomi regime. When Hidenaga's house ended, Izumi became a direct holding of the Toyotomi family, and following the Siege of Osaka, transferred to Tokugawa rule in 1615.

Miyoshi Nagayoshi founded Nanshūji to honor his father's memory, with Dairin Sōtō of Daitokuji serving as the founding abbot.

While touring the Kinai after visiting Azuchi, Tokugawa Ieyasu learned at Sakai of the deaths of Nobunaga and Nobutada and hurried to return to his domains with a small retinue.
